IL6 and TNF: The Inflammation Genes Behind a "Leaky" Gut

Two cytokines, one gut barrier

IL6 (interleukin-6) and TNF (tumor necrosis factor alpha) are two of the body's master inflammatory signals, involved in everything from fighting infection to healing wounds. That response can, in turn, further affect gut barrier function, a loop connecting diet, gut bacteria, gut permeability and inflammation that is now one of the more active areas of metabolic research.

Where genetic variation comes in

Variants in the regulatory regions of IL6 and TNF are associated with differences in how much of each cytokine someone produces in response to the same trigger. That does not mean a "high" variant causes gut problems, most people with these variants have no digestive symptoms at all, but it is one more piece of a very large picture that includes diet, sleep, stress, and exercise, all of which influence inflammatory tone more than any single gene does.
